Diets High in Animal Protein May Help Prevent Functional Decline in Elderly Individuals

NEW YORK—A diet high in protein, particularly animal protein, may help elderly individuals maintain a higher level of physical, psychological, and social function according to a study published in the Journal of the American Geriatrics Society.——Due to increasing life expectancies in many countries, increasing numbers of elderly people are living with functional decline, such as declines in cognitive ability and activities of daily living. This can have profound effects on the health and well-being of older adults and their caregivers, as well as on health care resources.–Research suggests that as people age, their ability to absorb or process protein may decline. To compensate for this loss, protein requirements may increase with age. Megumi Tsubota-Utsugi, PhD, MPH, RD, of the National Institute of Health and Nutrition in Japan, and her colleagues in Tohoku University and Teikyo University, Japan, wondered whether protein intake might affect the functional capabilities of older adults. They designed a study to investigate the relationship between protein intake and future decline in higher-level functional capacity in older community-dwelling adults in Japan. Their analysis included 1,007 individuals with an average age of 67.4 years who completed food questionnaires at the start of the study and seven years later. Participants were divided into four groups (quartiles) according to their intake levels of total, animal, and plant protein. Tests of higher-level functional capacity included social and intellectual aspects as well as measures related to activities of daily living.—Men in the highest quartile of animal protein intake had a 39 percent decreased chance of experiencing higher-level functional decline than those in the lowest quartile. These associations were not seen in women. No consistent association was observed between plant protein intake and future higher-level functional decline in either sex.–“Identifying nutritional factors that contribute to maintaining higher-level functional capacity is important for prevention of future deterioration of activities of daily living,” said Dr. Tsubota-Utsugi. “Along with other modifiable health behaviors, a diet rich in protein may help older adults maintain their functional capacity.”

FAO reports rise in GM material in non-GM food trade

The FAO said 75 out of 193 member countries responded to questions about low level GM crops found
in non-GM food and feed imports in the first survey of its kind, carried out from February to June last year.
They reported 198 cases of GM crops in non-GM deliveries from 2002 to 2012, with 70% of those (138) detected since 2009.
Linseed, maize, rice and papaya were the most common crops found to contain unauthorised GM material.
Food safety officer at the FAO, Sarah Cahill, said:“The biggest concern is the disruption to trade between countries and of course
the associated economic impact. When low levels of GM crops are detected in a shipment of non-GM crops, the
shipment is generally rejected by the importing country. The confinements are either returned to the country of
origin, which in itself has cost implications, or in some cases they are destroyed. Now, the latter not only has economic
implications but also contributes to food losses Under current EU regulations, the threshold limit for unintentional presence of
GM materials in crops is 0.9%. Above this level, items containing approved GM products require labelling.[F6]
However, most of the 75 countries involved in the study did not have policies in place for low levels of GM material. Cahill added
that with increased production of GM crops – and improved sensitivity and frequency of testing – it was probably not unusual that
detection was increasing in traded food. Consumer implications? Any of these GM products that are in the market have been subject
to rigorous safety assessments, so from a safety perspective they are not marketed without undergoing that level of assurance, Cahill
said.“We are aware that many consumers like to make a choice between GM foods and non-GM foods and, if it’s not clear whether a
non-GM food has low levels of GM foods, this may have implications for the choice that they make. They may not have all the information
they need to make that choice FAO senior food safety officer in charge of the survey Renata Clarke said the FAO also asked countries to
help the organisation assess the safety of GM crops for human consumption We would like to see countries sharing any scientific
findings they have on the subject, she said. With this in mind, the FAO has set up a GM foods platform for countries to share information
on safety assessments, The survey is one way in which the FAO aims to tackle the issue of low level unauthorised GM crops in shipments,
with the resulting information

OBJECTIVES: The K-ras gene mutation commonly found in lung adenocarcinomas contributes to their non-invasive expansion. Our main objective here was to develop a chemopreventive agent against K-ras-mutated lung adenocarcinoma cell line like-A549.
MATERIALS AND METHODS: We isolated flavonol from ethanolic leaf extract of Thuja occidentalis[F7], and evaluated its apoptotic potentials on A549 cells. They were treated with 1-10 μg/ml of flavonol and viability was tested retaining normal lung cells L-132 as control. We performed assays such as TUNEL, annexin V, cell-cycle and mitochondrial membrane potentials, by FACS analysis. ROS-mediated oxidative stress and drug-DNA interactions were analysed along with gene expression studies for p53, Bax-Bcl2, cytochrome c, the caspase cascade genes and PARP.
RESULTS: Flavonol reduced A549 cell viability in a dose- and time-dependent manner (IC50 value = 7.6 ± 0.05 μg/ml following 48 h incubation) sparing normal L-132 cells. It effected G2-M phase cell cycle arrest and apoptosis, as indicated by progressive increase in the sub-G1, annexin V and TUNEL-positive cell populations. Apoptotic effects appeared to be mitochondria-dependent, caspase-3-mediated, but ROS-independent. Analysis of circular dichroism data revealed that flavonol intercalated [F8]with nuclear DNA. In vivo studies on non small cell lung carcinoma (NSCLC)-induced mice confirmed anti-cancer potential of flavonol.
CONCLUSION: Flavonol-induced apoptosis apparently resulted from intercalation of cells’ nuclear DNA. Flavonol inhibited growth of induced lung tumours in the mice, indicating its potential as an effective agent against NSCLC.—PMID: 24267912 [PubMed – indexed for MEDLINE]

Soy infant formula and seizures in children with autism- a retrospective study

Seizures are a common phenotype in many neurodevelopmental disorders including fragile X syndrome, Down syndrome and autism. We hypothesized that phytoestrogens in soy-based infant formula were contributing to lower seizure threshold in these disorders. Herein, we evaluated the dependence of seizure incidence on infant formula in a population of autistic children. Medical record data were obtained on 1,949 autistic children from the SFARI Simplex Collection. An autism diagnosis was determined by scores on the ADI-R and ADOS exams. The database included data on infant formula use, seizure incidence, the specific type of seizure exhibited and IQ. Soy-based formula was utilized in 17.5% of the study population. Females comprised 13.4% of the subjects. There was a 2.6-fold higher rate of febrile seizures [4.2% versus 1.6%, OR = 2.6, 95% CI = 1.3-5.3], a 2.1-fold higher rate of epilepsy comorbidity [3.6% versus 1.7%, OR = 2.2, 95% CI = 1.1-4.7] and a 4-fold higher rate of simple partial seizures [1.2% versus 0.3%, OR = 4.8, 95% CI = 1.0-23] in the autistic children fed soy-based formula. No statistically significant associations were found with other outcomes including: IQ, age of seizure onset, infantile spasms and atonic, generalized tonic clonic, absence and complex partial seizures. Limitations of the study included: infant formula and seizure data were based on parental recall, there were significantly less female subjects, and there was lack of data regarding critical confounders such as the reasons the subjects used soy formula, age at which soy formula was initiated and the length of time on soy formula. Despite these limitations, our results suggest that the use of soy-based infant formula may be associated with febrile seizures in both genders and with a diagnosis of epilepsy in males in autistic children. Given the lack of data on critical confounders and the retrospective nature of the study, a prospective study is required to confirm the association.


Study Suggests Link Between Soy Formula and Seizures in Autistic Children

Madison, Wisconsin – A University of Wisconsin-Madison researcher has detected a higher rate of seizures among children with autism who were fed infant formula containing soy protein rather than milk protein.—“This needs to be studied more thoroughly. If soy formula is lowering the threshold for seizures or increasing the incidence of seizures, we need to know that.”- The study found excess seizures among girls and in the total sample of 1,949 children. The soy-seizure link reached borderline significance among boys, who comprised 87 percent of the children described in the database under study.—Seizures – caused by uncontrolled electrical currents in the brain – occur in many neurological disorders including epilepsy, Alzheimer’s disease, Down syndrome and autism.—About 25 percent of infant formula sold in the United States is based on soy protein.[F9] Study author Cara Westmark, a senior scientist in the Department of Neurology at the UW School of Medicine and Public Health, says her investigation was sparked by mouse studies of a drug that, it was hoped, would inhibit seizures by blocking signals that excite nerve cells. “It was pure serendipity that we happened to look at soy,” she says. To simplify the mouse study, she replaced the standard lab chow, which had a variable composition, with a diet containing purified ingredients. Unexpectedly, that diet reduced the rate of seizures by 50 percent compared to standard chow, Westmark says. “We were intrigued that a dietary alteration was as effective as many medicines in reducing seizure incidence and wanted to pursue that finding,” she says. “We found that the main difference between the diets was the protein source. The standard diet was soy-based, while the purified diet was casein, or dairy, based.”—The mechanism of action is unknown, but Westmark points to the high level of plant-derived estrogens in soy products as a possible cause of the excess seizures.

Prevalance of Seizures Small But Significant

People eat a lot of soy products, and when Westmark began to look for the effect in people, she decided to focus on infants, who may consume nothing but formula. Knowing that people with autism have a higher rate of seizures, Westmark turned to a database from the Simons Foundation Autism Research Initiative.—And that led to the new study, published today in the journal PLOS ONE, which showed that children with autism who were fed soy formula had 2.6 times as many febrile seizures as the children fed non-soy formula in the database. That means 4.2 percent of the soy group had a seizure associated with a fever, compared to 1.6 percent of the others.–To put it another way, the vast majority of both groups did not have seizures.—“This is not saying that all autistic children who eat soy-based formula are going to develop seizures,” says Westmark.–And yet that increase is worrying, Westmark says. –“The prevalence of autism is increasing and currently affects one American child in 88,” she says. “Soy is a widespread ingredient in many food products and 25 percent of infant formulas are soy based, so this is something that needs to be studied. If the child is lactose intolerant, there are alternatives that a pediatrician can recommend.”—-

The mechanisms behind autoimmune reaction to nervous system antigens in autism are not understood. We assessed the reactivity of sera from 50 autism patients and 50 healthy controls to specific peptides from gliadin and the cerebellum. A significant percentage of autism patients showed elevations in antibodies against gliadin and cerebellar peptides simultaneously. For examining cross-reaction between dietary proteins and cerebellar antigens, antibodies were prepared in rabbits, and binding of rabbit anti-gliadin, anti-cerebellar peptides, anti-MBP, anti-milk, anti-egg, anti-soy and anti-corn to either gliadin- or cerebellar-antigen-coated wells was measured. In comparison to anti-gliadin peptide binding to gliadin peptide at 100%, the reaction of anti-cerebellar peptide to gliadin peptide was 22%, whereas the binding of anti-myelin basic protein (MBP), anti-milk, anti-egg and anti-soy to gliadin was less than 10%. Further examination of rabbit anti-gliadin (EQVPLVQQ) and anti-cerebellar (EDVPLLED) 8 amino acid (AA) peptides with human serum albumin (HSA) and an unrelated peptide showed no binding, but the reaction of these antibodies with both the cerebellar and gliadin peptides was greater than 60%. This cross-reaction was further confirmed by DOT-immunoblot and inhibition studies. We conclude that a subgroup of patients with autism produce antibodies against Purkinje cells and gliadin peptides, which may be responsible for some of the neurological symptoms in autism.
